Semaglutide is a GLP-1 receptor agonist built on the native GLP-1(7-36) sequence with two modifications that define its pharmacology: alpha-aminoisobutyric acid replaces alanine at position 2, which blocks dipeptidyl peptidase-4 cleavage, and lysine-26 carries a C18 fatty diacid on a glutamic acid linker that binds serum albumin. Molecular formula C187H291N45O59, molecular weight 4113.6 g/mol. Lau and colleagues (Journal of Medicinal Chemistry, 2015) reported that the C18 diacid raised albumin affinity roughly 4.8-fold over the C16 palmitoyl chain used in liraglutide, the structural basis for its roughly one-week half-life in preclinical pharmacokinetic models. Research Summary

The GLP-1 receptor is a class B G protein-coupled receptor expressed on pancreatic beta cells, in the hypothalamus and the brainstem nucleus tractus solitarius, and in gastric and cardiovascular tissue. Agonist binding couples to G-alpha-s, raising cyclic AMP and activating protein kinase A and Epac2. In isolated islet preparations this potentiates glucose-stimulated insulin secretion only above a glucose threshold, which is why the in-vitro literature describes the effect as glucose-dependent.

Animal-model work has examined semaglutide in hypothalamic appetite circuits, gastric emptying, beta-arrestin recruitment kinetics and cardiovascular biomarkers. For a laboratory the analytical questions matter as much as the pharmacology. The acylated lysine and the Aib substitution mean identity has to be confirmed by mass spectrometry against the expected mass rather than by retention time alone, and the lipidated side chain makes the compound prone to aggregation in solution. Storage and Handling
Store the sealed lyophilised vial at minus 20 degrees Celsius for long-term preservation, or 2 to 8 degrees Celsius for short-term storage. After reconstitution, store at 2 to 8 degrees Celsius and protect from light. Allow vials to equilibrate to room temperature before opening to limit condensation.
