Research Summary

5-Amino-1MQ (5-Amino-1-methylquinolinium) is a small molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that catalyzes the methylation of nicotinamide using S-adenosylmethionine (SAM) as a methyl donor. NNMT has been identified as a key metabolic regulator in adipose tissue, and its overexpression has been observed in preclinical obesity models. By inhibiting NNMT, 5-Amino-1MQ has been investigated for its effects on cellular NAD+ levels, SAM availability, and downstream metabolic pathway regulation.

In-vitro studies using 3T3-L1 adipocyte cultures have observed that NNMT inhibition by 5-Amino-1MQ reduces lipid accumulation, decreases adipocyte size, and modulates the expression of adipogenic transcription factors including PPARγ and C/EBPα. Animal model research in diet-induced obesity rodent models has investigated the effects of 5-Amino-1MQ on body weight, adipose tissue mass, cholesterol levels, and metabolic rate parameters. Preclinical findings indicate that NNMT inhibition influences the intracellular methylation balance and NAD+ salvage pathway flux.

Additional preclinical research has explored the role of NNMT in cellular energy metabolism beyond adipose tissue, including studies in muscle cell cultures where NNMT inhibition has been observed to increase NAD+-dependent sirtuin activity and modulate mitochondrial biogenesis markers. The cell-permeable properties and selectivity profile of 5-Amino-1MQ have been characterized in competitive binding and enzyme kinetics assays.
