GHRP-2 (pralmorelin) is a synthetic hexapeptide agonist at the growth hormone secretagogue receptor (GHS-R1a), sequence D-Ala-D-2-Nal-Ala-Trp-D-Phe-Lys-NH2, molecular formula C45H55N9O6, molecular weight 817.97 g/mol. Two D-amino acids at positions 1 and 5 and a C-terminal amide protect the chain from aminopeptidase and carboxypeptidase cleavage, which is why it outlasts endogenous ghrelin in preclinical assays. Bowers and colleagues developed it in the early 1990s as a second-generation analogue of GHRP-6, with D-2-naphthylalanine at position 2 as the pharmacophore that drives receptor affinity. Research Summary

GHS-R1a is a Gq/11-coupled receptor on anterior pituitary somatotrophs and in the hypothalamic arcuate nucleus. Receptor occupancy activates phospholipase C, generating inositol trisphosphate and diacylglycerol, and the resulting calcium release drives exocytosis of GH-containing secretory granules. In bovine pituitary cell cultures, Cheng and colleagues (Journal of Endocrinology, 1997) showed that GHRP-2 also recruits protein kinase C and cyclic AMP signalling, a partial overlap with the GHRH receptor pathway that accounts for the amplified GH output seen when the two receptor classes are engaged together in research models.

Beyond the pituitary, GHRP-2 binds the CD36 scavenger receptor, and preclinical work has examined GH-independent activity in hepatic, cardiac and musculoskeletal tissue. Granado and colleagues (2005) reported a lower arthritis score in rats with adjuvant-induced arthritis. In most laboratories GHRP-2 serves as the reference GHS-R1a agonist against which newer, more selective secretagogues such as ipamorelin are benchmarked. Store the sealed lyophilised vial at minus 20 degrees Celsius for long-term preservation, or 2 to 8 degrees Celsius for short-term storage. After reconstitution, store at 2 to 8 degrees Celsius and protect from light.
