Research Summary

SS-31 (Elamipretide) is a mitochondria-targeting tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH₂. Its unique alternating aromatic-cationic motif enables selective accumulation in the inner mitochondrial membrane, where it has been investigated for its interaction with cardiolipin. Cardiolipin is a phospholipid critical for electron transport chain (ETC) complex organization, and preclinical research suggests SS-31 stabilizes cardiolipin-dependent cristae structure and optimizes electron transfer efficiency.

In animal models of ischemia-reperfusion injury, SS-31 has been studied for its effects on mitochondrial reactive oxygen species (ROS) production, cytochrome c release, and mitochondrial membrane potential. In-vitro findings indicate that SS-31 reduces mitochondrial superoxide generation without acting as a direct free radical scavenger, instead modulating the efficiency of electron flow through Complex III and Complex IV.
