Research Summary
SS-31 (Elamipretide) is a mitochondria-targeting tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH₂. Its unique alternating aromatic-cationic motif enables selective accumulation in the inner mitochondrial membrane, where it has been investigated for its interaction with cardiolipin. Cardiolipin is a phospholipid critical for electron transport chain (ETC) complex organization, and preclinical research suggests SS-31 stabilizes cardiolipin-dependent cristae structure and optimizes electron transfer efficiency.
In animal models of ischemia-reperfusion injury, SS-31 has been studied for its effects on mitochondrial reactive oxygen species (ROS) production, cytochrome c release, and mitochondrial membrane potential. In-vitro findings indicate that SS-31 reduces mitochondrial superoxide generation without acting as a direct free radical scavenger, instead modulating the efficiency of electron flow through Complex III and Complex IV. Research in rodent models has also examined its influence on ATP synthesis rates and mitochondrial bioenergetics under metabolic stress conditions.
Preclinical investigations in aged animal models have explored the relationship between SS-31 and age-related mitochondrial dysfunction, including studies measuring skeletal muscle energetics, cardiac function parameters, and renal mitochondrial morphology. Its favorable cell permeability and mitochondrial concentration properties have been characterized in pharmacokinetic studies across multiple species.
