5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule, membrane-permeable inhibitor of nicotinamide N-methyltransferase (NNMT), the cytosolic enzyme that transfers a methyl group from S-adenosylmethionine to nicotinamide to form 1-methylnicotinamide. It is not a peptide, but it is catalogued alongside research peptides because the NNMT reaction sits at the junction of NAD+ salvage and cellular methylation, the same axis studied with NAD+ and MOTS-c. Research Summary

NNMT overexpression drains two substrates at once: nicotinamide, the precursor for NAD+ synthesis through the NAMPT salvage pathway, and S-adenosylmethionine, the universal methyl donor. Kraus and colleagues (Nature, 2014) showed that antisense knockdown of NNMT in the white adipose tissue of diet-induced obese mice reduced adiposity and raised both SAM and NAD+ in fat tissue, which established the enzyme as a metabolic research target. 5-Amino-1MQ mimics the quaternary nitrogen of 1-methylnicotinamide and competes for the substrate pocket, so it inhibits the enzyme without genetic manipulation.

Neelakantan and colleagues at the University of Texas Medical Branch (Biochemical Pharmacology, 2018) characterised the compound in 3T3-L1 adipocytes, where treatment lowered intracellular 1-methylnicotinamide, raised NAD+ by 1.2 to 1.6 fold and suppressed lipogenic gene expression, and in diet-induced obese mice, where it reduced white adipose tissue mass and improved plasma lipid profiles without changing food intake. Because it is a quinolinium salt rather than a peptide, its analytical profile differs from the rest of the catalogue: there is no peptide content or amino acid analysis to report, and purity rests on HPLC with mass confirmation. Storage and Handling
Store the sealed lyophilised vial at minus 20 degrees Celsius for long-term preservation, or 2 to 8 degrees Celsius for short-term storage.
